How Stiffness Affects Cell Senescence and Their Secretions

Did you know that the stiffness of the environment where cells live can change how they age and what they release? This is like how a rubber band feels different on a soft pillow compared to a hard surface. In a condition like pulmonary fibrosis, the tissue gets stiffer and more senescent cells appear. Senescent cells release a special mix of proteins called a senescence-associated secretory phenotype (SASP). Scientists wanted to see how this stiffness affects the SASP. They used human lung cells and grew them on different surfaces, from soft to very hard.

They then added a drug called doxorubicin to make the cells senescent. The results? Stiffer surfaces made the cells release different proteins, especially more collagen. This was seen not just in the lab but also in mice with lung fibrosis and in human lung disease data. It turns out, the environment's stiffness really shapes how cells age and what they release.
