How to Best Preserve Mouse Testes for Research

Have you ever wondered how scientists keep mouse testes fresh for their studies? It turns out, the way they're stored makes a big difference. This research compared different methods of preserving mouse testes. They used two types of buffers, RNAlater® and Nucleic Acid Preservation-NAP-buffer, and stored the samples at various temperatures like room temperature, 4°C, -20°C, -80°C, and even liquid nitrogen. The goal? To see which method kept the genetic material, called RNA, in the best shape. The study showed that storing the testes in RNAlater® or NAP buffer at -80°C was as good as keeping somatic tissues (like skin or muscle) fresh. When kept at room temperature for a week, both buffers did a great job. But if you store them longer, like a year, you start to see a drop in quality.

Interestingly, samples stored at 4°C and -20°C held up well too. Even after a year, the RNA quality and protamine expression, which are important for studying fertility, remained stable. The best news? Snap-freezing in liquid nitrogen and then storing at -80°C or -196°C worked super well.
