Small Blood, Big Clues: Fingerprinting Animal Species

The world is full of pets and wildlife that sometimes find their way into our homes. When an animal gets hurt or causes trouble, detectives need a quick way to tell which species left the blood behind.

Scientists have made progress by using a technique called peptide mass fingerprinting (PMF). The old method needed a lot of blood and only worked well with mammals.

To fix this, researchers switched to an “in‑gel digestion” version of PMF. They first ran mixed blood samples from ten different animals on an SDS‑PAGE gel, then used software called Mascot to spot the proteins.

After finding the best amount of protein for the test, they ran the same gel‑digestion PMF on blood from 21 different species and on bloodstains from 10 species.

In almost every case, the hemoglobin protein from the right animal—or a closely related one—scored highest. This means the new method can guess the species from even tiny drops of blood.

Because it only needs a small sample and works with an inexpensive single mass spectrometer, this approach could make forensic protein testing easier for many labs.

The study shows that by tweaking the procedure, scientists can turn a small blood stain into useful evidence about which animal was involved.
